Intimal smooth muscle cells: the context-dependent origin.

چکیده
The sixth-century BC Chinese military general Sun Tzu underscored the dangers of overconfidence in strategy. A similar view has been shared by other thinkers and applied further to various contexts other than the military. A major goal of the medical sciences is to develop effective, wellrefined therapies. Accomplishing this task requires a comprehensive understanding of the biology of each component in the disease mechanism and realization of the limits of current knowledge. This view can apply not only to the development of new therapies but also to advanced diagnostics (eg, imaging and biomarkers).

The origins of atherosclerosis.
The presence of atherosclerotic lesions in young adults suggests that early stages of atherogenesis occur during childhood. The relationship of intimal lesions in childhood to fully developed atherosclerosis is briefly discussed. Factors likely to promote lipid accumulation within the arterial wall and proliferation of connective tissue elements are reviewed with particular emphasis on endothe...
متن کاملNeointima formation: a local affair.
Where do intimal smooth muscle cells (SMCs) come from? For many years, the idea that intimal SMCs originated from the underlying media went unchallenged.1 Then, reports2,3 began to appear that up to half of the SMCs in the intima of atherosclerotic plaques and injured arteries arose from circulating progenitor cells of bone marrow origin. This new view of intimal SMC formation was potentially i...

متن کاملContributions to the study of the foetal development of physiological intimal thickening in the human uterine artery.
The morphological study of the development of "intimal thickenings" of the human uterine artery in physiological condition was performed on 72 uterine arteries obtained from foetuses from the 12th week of gestation up to birth. Our results indicate that intimal thickening is formed by the migration and displacement of mesenchymal cells around the site of origin of collateral vessel from uterine...
متن کاملMechanisms of TGF-beta1-induced intimal growth: plasminogen-independent activities of plasminogen activator inhibitor-1 and heterogeneous origin of intimal cells.
Transforming growth factor (TGF)-beta(1) is a potent stimulator of intimal growth. We showed previously that TGF-beta(1) stimulates intimal growth through early upregulation of plasminogen activator inhibitor-1 (PAI-1) and, subsequently, PAI-1-dependent increases in cell migration and matrix accumulation. We also showed that PAI-1 negatively regulates TGF-beta(1) expression in the artery wall. ...
